Palvella Receives Priority Review for QTORIN Rapamycin

The FDA set a February 28, 2027 PDUFA date after accepting the company’s NDA, while third-quarter net loss widened to $24.6 million.

Palvella Therapeutics (PVLA), a clinical-stage biopharmaceutical company developing therapies for rare skin diseases and vascular anomalies, said the FDA accepted its new-drug application for QTORIN™ rapamycin in microcystic lymphatic malformations and granted Priority Review, with a February 28, 2027 PDUFA target action date.

The acceptance completed a rolling submission that had, in the prior quarter, been only at its first module, with full NDA completion still on track for the second half of 2026. Founder and Chief Executive Officer Wes Kaupinen said the company believes it is closer than ever to bringing its first therapy to patients.

The company said it is advancing preparations for a standalone U.S. commercial launch in the first half of 2027, if approved, with commercial, medical, and patient-services capabilities in place. It also received a third year of FDA Office of Orphan Products Development grant funding to support the Phase 3 SELVA trial and an open-label extension. QTORIN™ rapamycin could become the first FDA-approved therapy for microcystic lymphatic malformations, a disease affecting an estimated more than 30,000 diagnosed children and adults in the United States.

Research and development expenses were $10.8 million for the three months ended September 30, 2026, compared with $6.5 million a year earlier. The company attributed the increase to manufacturing, clinical development of QTORIN™ rapamycin for angiokeratomas, costs of completing the rolling NDA, and higher headcount and consulting. General and administrative expenses were $11.9 million, versus $3.6 million a year earlier, which the company attributed to higher headcount and professional services related to operating as a public company.

Net loss was $24.6 million, or $1.70 a share, for the three months ended September 30, 2026, compared with a net loss of $11.3 million, or $1.03 a share, a year earlier. Weighted average shares outstanding were 14,531,595 in the quarter. Shares outstanding were 15,844,715 as of November 2, 2026, including common share equivalents from pre-funded warrants.

Cash, cash equivalents, and short-term investments were $236.7 million as of September 30, 2026, down from $250.6 million as of June 30, 2026.

Beyond the lead program, Palvella said Phase 3 initiation for QTORIN™ rapamycin in cutaneous venous malformations remains on track for the fourth quarter of 2026. Enrollment was underway in LOTU, a Phase 2 trial of Fast Track-designated QTORIN™ rapamycin for clinically significant angiokeratomas, with topline results still expected in the second half of 2027. Phase 2 initiation of QTORIN™ pitavastatin for disseminated superficial actinic porokeratosis is expected in the fourth quarter of 2026, a tighter window than the second-half-of-2026 plan given in the two prior quarterly updates.

The company said it plans to announce a fourth QTORIN™ rapamycin indication and a third QTORIN™ product candidate in the fourth quarter of 2026. James R. Treat, M.D., a pediatric dermatologist and prior SELVA investigator, was appointed Chief Medical Officer effective December 21, 2026.

Kaupinen said the company’s focus is to obtain FDA approval, bring its first therapy to patients, advance additional QTORIN™ programs, and build a company addressing serious, rare skin diseases and vascular anomalies.
