Cogent Widens Third-Quarter Loss as Launch Costs Rise
General and administrative expenses reached $42.1 million in the three months ended September 30, 2026, as the company prepared for a November 30 PDUFA date on bezuclastinib in GIST.
Cogent Biosciences (COGT), a biotechnology company developing precision therapies for genetically defined diseases, reported a net loss of $111.4 million for the third quarter of 2026, compared with a net loss of $80.9 million a year earlier, as it built a commercial organization ahead of a potential bezuclastinib launch.
President and Chief Executive Officer Andrew Robbins said the commercial team is in place and the company is prepared to execute on the anticipated launch of bezuclastinib in gastrointestinal stromal tumors, with a PDUFA action date of November 30, 2026.
General and administrative expenses were $42.1 million in the three months ended September 30, 2026, up from $14.4 million in the third quarter of 2025. The company said the increase was primarily due to continued investments in commercial readiness, including personnel and infrastructure to support the potential launch of bezuclastinib, as well as overall organizational growth. Non-cash stock compensation in G&A was $9.4 million, compared with $5.2 million a year earlier.
Research and development expenses were $75.1 million, up from $69.0 million in the third quarter of 2025. The company attributed the increase primarily to costs incurred to support the ongoing SUMMIT, PEAK and APEX clinical trials and to the continued progression of early-stage, preclinical and discovery programs. Non-cash stock compensation in R&D was $7.5 million, versus $5.4 million a year earlier.
Total operating expenses were $117.2 million in the three months ended September 30, 2026, versus $83.4 million in the three months ended September 30, 2025. Interest income was $7.1 million, compared with $3.9 million a year earlier.
Cash, cash equivalents and marketable securities were $791.1 million as of September 30, 2026, down from $792.3 million as of June 30, 2026. The company said the balance is expected to fund operations into late 2028, well past all three pending PDUFA action dates for bezuclastinib.
The FDA accepted the New Drug Application for bezuclastinib in Advanced Systemic Mastocytosis, with a PDUFA target action date of June 29, 2027. The company had submitted that NDA on June 30 in the prior quarter. A NonAdvSM PDUFA target action date is December 30, 2026.
Cogent said it expects to file INDs this quarter for CGT1815, a selective pan-KRAS(ON) inhibitor, and CGT1145, a selective JAK2 V617F inhibitor, and to complete dose escalation for CGT4255, a CNS-penetrant mutant ErbB2 inhibitor.
The company also announced the initiation of a single-arm, 40-patient extension cohort of the PEAK trial investigating bezuclastinib in combination with sunitinib in first-line GIST patients, designed to prospectively measure ORR and PFS in patients with either exon 9 or exon 11 primary mutations. It plans to present multiple posters featuring bezuclastinib at the 68th Annual Meeting of the American Society of Hematology, December 12–15, 2026, in New Orleans, including SUMMIT analyses of skin and neurocognitive symptoms and the first look at clinical results from a SUMMIT sub-study of switching from avapritinib to bezuclastinib.
