Lexeo Starts LX2006 Pivotal Trial, Wins RMAT for LX2020

Cash, cash equivalents and investments were $234.2 million as of June 30, 2026, which the company said it expects to fund operations into 2028.

Lexeo Therapeutics (LXEO), a clinical-stage genetic medicine company focused on cardiovascular disease, initiated SUNRISE-FA 2 in June after finalizing the open-label pivotal protocol and statistical analysis plan for LX2006 in Friedreich ataxia cardiomyopathy. The first trial site was activated, and the first patient was enrolled.

The start matched the company’s plan to begin the study in the second quarter of 2026. In the first quarter, it said the protocol and SAP had been submitted to the FDA and that it was awaiting final feedback. Site activation is ongoing.

Chief Executive Officer R. Nolan Townsend said the company closed the quarter with momentum and that, with SUNRISE-FA 2 underway, it is focused on execution and on what it believes could be the first disease-modifying gene therapy for FA cardiomyopathy.

CLARITY-FA, a natural-history study, continues enrollment across sites in the United States and globally. Patients enrolled in CLARITY-FA are eligible to join SUNRISE-FA 2, which the company said supports continued enrollment once trial sites are active.

Anticipated LX2006 milestones include continued FDA engagement on a confirmatory evidence strategy, with an update expected once that strategy is finalized; a topline data readout in the second half of 2027; and a potential BLA submission under the accelerated approval pathway in the first half of 2028.

In June, the company also announced publication of LX2006 data in JAMA Cardiology showing the candidate is generally well tolerated, with signs of efficacy in Phase I/II studies. In April, it presented data at ASGCT on the clinical profile and on preclinical and manufacturing work supporting potential sequential dosing.

The second clinical program advanced with a regulatory designation. In August, the FDA granted Regenerative Medicine Advanced Therapy designation for LX2020 in PKP2 arrhythmogenic cardiomyopathy, based on interim data from an ongoing Phase I/II trial. RMAT provides opportunities for increased FDA interaction and may help expedite development. LX2020 remains generally well tolerated across ten participants dosed, with no clinically significant complement activation to date.

Anticipated LX2020 milestones include a 12-month data update for all high-dose participants in the fourth quarter of 2026 and regulatory engagement with the FDA expected in 2026.

In May, the company presented preclinical data for LX2022 at ASGCT, showing proof-of-concept efficacy for TNNI3 gene replacement in a newly developed porcine model of hypertrophic cardiomyopathy.

Research and development expenses were $19.0 million for the three months ended June 30, 2026, compared with $14.7 million a year earlier. General and administrative expenses were $9.0 million, compared with $16.0 million a year earlier.

Net loss was $25.9 million, or $0.30 a share, basic and diluted, for the three months ended June 30, 2026, compared with $26.1 million, or $0.60 a share, a year earlier. Weighted-average shares used in the per-share calculation were 85.6 million.

Cash, cash equivalents, and investments in marketable securities were $234.2 million as of June 30, 2026. That amount is expected to fund operations into 2028.

Townsend said the company has multiple value-driving catalysts ahead and a strong balance sheet to support key clinical and regulatory milestones.
